Abstract

Instructions: The objective of these studies is to determine the kinetics of ceftazidime hydrolysis in the pH range of 5 to 6. This is the region of maximum stability. The ultimate goal is to formulate solution dosage forms at concentrations as high as 0.5 to 1 M ceftazidime. These studies will be conducted at 65 C. Ceftazidime has three pKa values: carboxylic acid pKa values at 1.9 and 2.7 and aminothiazolyl pka at 4.1. There are no salt effects and buffer component concentrations need only by 5X greater than the initial substrate concentration to maintain constant pH. Although the degradation scheme is complex, you should attempt to explain any kinetic phenomena in terms of the chemistry of the products and substrates.
